5-Amino-1MQ (5-amino-1-methylquinolinium) is a small molecule inhibitor of NNMT (nicotinamide N-methyltransferase), an enzyme that catalyzes the methylation of nicotinamide using S-adenosyl methionine (SAM) as the methyl donor. NNMT is overexpressed in adipose tissue and various cancer cell lines, and its inhibition has been studied as a strategy to increase intracellular NAD+ levels, enhance one-carbon metabolism, and reduce adipogenesis in preclinical models. 5-Amino-1MQ was identified as a potent NNMT inhibitor in work published by the Bhatt Lab and collaborators, with subsequent preclinical studies demonstrating that NNMT inhibition in diet-induced obese mice reduced body weight, decreased adipose tissue mass, and improved metabolic parameters including insulin sensitivity and plasma lipids. The proposed mechanism involves NNMT inhibition increasing the SAM:SAH ratio and NAD+ precursor availability, activating sirtuin-mediated metabolic programs. Research interest has also extended to NNMT’s role in cancer metabolism and tumor microenvironment modulation, where its overexpression is associated with poor prognosis in multiple cancer types.
